چکیده

24 Vibrio cholerae serotype O1 causes diarrhea in many developing countries around the world. In most cases, cholera patients pose some sorts of immunity against the naturally occurring infection. In the present study, the bactericidal activity of the sera of rabbit immunized with whole cell extract of Vibrio cholerae O1 was investigated. The protein profile of the serotype exhibited 20 types of proteins of distinguished molecular weights. The western blot analysis revealed at least 10 proteins to be immunogenic. Interestingly, decomplementation of the sera was found to abolish the bactericidal activity. The bactericidal activity of non immunized rabbit sera was also investigated and the results revealed that the immunized sera were more effective than that of non immunized one. Production of Vibrio cholerae O1 and non-O1 typing sera in rabbits immunized with polysaccharide-protein carrier conjugates.

Two systems are currently used to serologically type Vibrio cholerae O1 and non-O1 isolates. Antiserovar-serotype serum in the Smith system is produced in rabbits immunized with live whole-cell vaccines, and that in the Sakazaki system is produced in rabbits immunized with heat-killed vaccines. In neither system is the serovar-serotype-specific antigen clearly defined. During the course of a se...

Designing and Expression of Recombinant Chimeric Protein Containing CtxB and OmpW from Vibrio Cholerae and Evaluation of Its Immunogenicity

Background: Cholera disease caused by Vibrio cholerae remains a major cause of morbidity and mortality throughout the world. Various strategies with different proteins as immunogens have been tried for vaccine development, none of which have been sufficiently effective to preclude cholera. Chimeric proteins, with their ability to present multiple antigens at the same time, can play important ro...

Enterotoxin-specific immunoglobulin E responses in humans after infection or vaccination with diarrhea-causing enteropathogens.

Cholera toxin (CT)-specific antibody responses of the immunoglobulin E (IgE) isotype in the sera of adult patients suffering from infection with either Vibrio cholerae O1, V. cholerae O139, or enterotoxigenic Escherichia coli (ETEC) were analyzed and compared with those in the sera of volunteers immunized with a bivalent B subunit O1/O139 whole-cell cholera vaccine. A significant IgE response t...

Rapid Screening of Toxigenic Vibrio cholerae O1 Strains from South Iran by PCR-ELISA

Background: The ability to sensitively detect Vibrio cholera with PCR-ELISA method represents a considerable advancement over alternative more time-consuming methods for detection of this pathogen. The aim of this research is to evaluate the suitability of a PCR-enzyme-linked immunosorbent assay for sensitive and rapid detection of V. cholera O1. Methods: The 398-bp sequence of a gene that cod...

Detection of Vibrio cholerae with monoclonal antibodies specific for serovar O1 lipopolysaccharide.

Six hybridoma cell lines, each of which produced a monoclonal antibody (MAb) against Vibrio cholerae O1 lipopolysaccharide (LPS), were established. Each MAb was active serologically by both enzyme-linked immunosorbent assay (ELISA) and the slide agglutination test. In the ELISA, each MAb was tested against 7 O1 and 9 non-O1 LPS preparations. Three MAbs reacted with both Inaba and Ogawa serovars...
